Research Topics:
The focus of our group is on (i) the development and application of DNA-Microarrays (“E. coli Pathoarray”) to determine the distribution of virulence genes in freshly collected clinical E. coli isolates from patients suffering urinary tract infection (UTI) and (ii) in vivo and in vitro transcriptome analyzes (quantitative RT-PCR; TaqMan®) of virulence factors of extraintestinal pathogenic E. coli (ExPEC) using a mouse infection model of ascending UTI. Working-packages of the past: Identification of novel virulence genes and virulence-associated DNA-fragments of ExPECs by means of suppressive-subtractive cloning using total genomic DNA-preparation of different clinical ExPEC isolates Development of a DNA-Microarray by spotting PCR-products of well-known and putative virulence factor-genes on glas slides. Genotyping of large collections of clinical ExPEC-isolates for the presence of different virulence factor genes leading to figure out a clearer picture of of ExPEC virulence and to obtain the tools for an individual risk assessment for each isolate. Expression profiling of selected virulence genes of ExPECs to gain insight into the regulation of virulence factors in vitro and in vivo The aim of our work is to improve our understanding of the infection process in UTI due to E. coli and to provide us with knowledge of in vivo expressed E. coli (virulence) genes, that might serve as further targets for therapy and prevention of this widespread infectious disease
