Research Topics:
The main research topics of our group concern studies on adhesion and growth in biofilm of L. monocytogenes on abiotic surfaces, the characterization of biodiversity to assess the risk linked to L. monocytogenes, the stress adaptation and survival. Our research works are focused on (i) the characterisation of genes/proteins involved in adhesion and biofilm formation, (ii) the characterisation of subproteomes (intracellular, surface and secreted proteins), (iii) the study of biodiversity and the comparison of clinical, asymptomatic carriage and food plants strains of L. monocytogenes strains, (iv) the adaptative response to environmental variations and technological stresses (temperature, salt, alkaline and biocide treatments) and (v) the effects of selected decontamination processes on surfaces of food plants. For the characterization of sessile mode of growth, different models have been developed to form biofilms either on stainless steel, PTFE or glass fibers. Different approaches are used for these studies: the characterisation of physico-chemical properties of bacterial surfaces; the observation by scanning electron microscopy or fluorescence microscopy of the biofilm formation; the assessment of (i) adhesion ability and biofilm formation and (ii) the efficiency of selected cleaning disinfection processes (with Biofilm Control Sas startup); the characterization of genes of interest; the proteome and traductome characterization and variations by 2-D SDS-PAGE, the image analysis of 2-D gels (ImageMaster 2-D Platinium, GE Healthcare) and the identification of proteins of interest by mass spectrometry (Maldi-Tof MS or ESI-Ion Trap MS, INRA Proteomic platform, responsible : M. Hébraud).
